Description: AmCyan-tagged F. graminearum PH-1 strain were used for inoculation of wheat seedling coleoptiles. Fungal hyphae at 16 hours post inoculation (hpi), 40 hpi and 64 hpi were isolated from infected tissues using laser microdissection (3 biological replicates each time point, sample name 16 hpi, 40 hpi and 64 hpi). Infected coleoptile tissues at 240 hpi were also collected (two biological replicates, smaple name 240 hpi). Macroconida (2 days cultured in liquid mungbean media) that used for inoculated were also sampled as control (sample name 0 hpi). Fungal hyphae after 72 hours cultured in YEPD liquid medium were collected as in vitro control (sample name 72 h). All the RNA smaple were linearly amplified two rounds (T7 based method) before chip hybridization.
