Description: One ml suspensions of 108 macroconidia of F. graminearum strain NRRL 31084 (PH-1) were inoculated into three 250 ml flasks containing 100 ml CM (Harris et al., 1994) and cultivated at 25 °C for 24 hours with shaking at 150 rpm. Mycelia of each flask were harvested and washed with sterile distilled water. The harvested mycelia were re-inoculated in 100 ml CM (complete media control), 100 ml minimal media prepared without NaNO3 (nitrogen starvation), and in 100 ml minimal media without dextrose (carbon starvation), respectively. After 12 hours of cultivation at 25 °C with shaking at 150 rpm, mycelia were harvested and used for RNA extraction.
