Description: Mutant cch1-14 (gene fg01364 (homologous to Saccharomyces cerevisiae CCH1 voltage-gated calcium channel YGR217w)was replaced with hygromycin phosphotransferase gene hph from pCB1004) in Fusarium graminearum PH-1 background inoculated on carrot agar, grown for five days, and sexual development was induced. Total RNA was harvested at time of induction (0H), times equivalent to wild-type 96H (croziers and ascus initials forming) and wild-type 144H (mature perithecia with multiseptate ascosopres), and labeled cRNA was hybridized to Fusariuma Affy GeneChips to detect differential expression during sexual development.
